Diflubenzuron is dissolved in 1,4-dioxane, separated by LC, and determined from peak areas vs linuron internal standard. Identity is confirmed by retention time.

Dissolve, in 100 mL volumetric flask, 4.2 mg diflubenzuron and 1.8 mg 1,3-di(4-chlorophenyl)urea in 20 mL dioxane, dilute to volume with solvent mixture, (b), and mix. Filter through 1.2 µm filter before injection.
Inject 20 µL onto column and determine resolution (R) with the following formula:

where R = resolution; d = distance between peak maxima; W 1 and W 2 = peak width at baseline of diflubenzuron and 1,3-di(4-chlorophenyl)urea, respectively. Resolution should be >1. If necessary, resolution can be improved by slightly increasing water content of mobile phase.

Determination
Inject 20 µL diflubenzuron standard solution, (d). Determine peak areas (or peak heights × retention times) of diflubenzuron and internal standard. Repeat injections until response ratio (area diflubenzuron peak/area internal standard peak) varies <1% for successive injections. Inject 20 µL test solution.
