Weigh 4–5 g test portion by difference into 250 mL centrifuge bottle and extract twice with 50 mL portions petroleum ether, shaking 5 min each time. Wash down sides of bottle with petroleum ether, centrifuge, and pour off solvent, disregarding opalescence. Warm bottle to drive off remaining solvent, transfer residue to mortar, and grind. Return fine powder to bottle with aid of 100 mL 10% NaCl solution (w/v). Shake bottle 15 min, wash down sides with NaCl solution, centrifuge well, and pour off supernate, disregarding opalescence. Repeat washing twice.
Extract in same manner, once with 70% alcohol (v/v) and once with H2O, shaking 1–2 min each time. Drain bottle several minutes, chill, and from pipet add 100 mL dilute HCl (20.5–21.0 g HCl/100 mL) at temperature ≤15°C. Shake vigorously 3 min, centrifuge well, and pour off solution through cotton pledget in funnel stem. Cool solution to temperature at which the HCl was added, and pipet off 50 mL into nursing bottle containing 115 mL alcohol. Shake with whirling motion 1 min, let stand 2 min, centrifuge 2 min, pour off through weighed Gooch containing glass fiber filter, and add 50 mL 70% alcohol (v/v) to precipitate. Stopper bottle, shake vigorously, wash down sides with the 70% alcohol (v/v), centrifuge lightly, and pour off through crucible. Repeat once with 70% alcohol (v/v) and once with alcohol. Dry crucible and contents 1.5 h at 130°C in air, or 5 h at 98°–100°C in vacuo. Cover crucible, place in desiccator containing efficient desiccant, and weigh when crucible reaches room temperature.
