Plates
Before assay, determine by preparation of trial plates optimum concentration (usually 0.05–0.2%) of organism suspension of Micrococcus luteus, 957.23D(e )(1) (see 5.3.01), to be added to agar medium J, 957.23A(j) (see 5.3.01), to obtain zones of inhibition of adequate size (17.5 mm ± 10% with reference concentration) and sharpness. For actual assay add appropriate amount suspension to agar medium J previously melted and cooled to 48°C. Place 10 mL inoculated medium in each of required number of plates, let harden, and refrigerate in inverted position until just before use.
Assay Solution
Accurately weigh ca 10 g test sample containing equivalent of ≥92.5 g erythromycin base/907 kg or 40 g containing equivalent of 9.25 g erythromycin base/907 kg and transfer to 250 mL glass-stoppered Erlenmeyer.
