A combination of thin layer and gas chromatographic methods for measuring urinary testosterone and epitestosterone glucuronide was employed using C–1, 2–3Htestosterone as a tracer. Values for combined testosterone and epitestosterone glucuronide in 24-hr urine specimens were: 65–348 μg for normal males aged 19–29; 45–178 μg for normal males aged 30–40; and 2–10 jug for normal females aged 10–53 years. Significant increases in urinary values were found in 13 of 15 subjects with “Stein-Leventhai” syndrome and in 5 of 9 patients with “idiopathic” hirsutism. A correlation between elevated urinary testosterone and epitestosterone levels and clinical manifestations of androgenicity were observed in subjects with the adrenogenital syndrome, Cushing's syndrome, virilizing ovarian neoplasm and acromegaly. Two subjects with anorchia excreted values similar to those of normal adult females.
