Antigenic thyroid substances have been investigated by agar diffusion techniques. Human thyroid antibodies obtained experimentally in the rabbit by the immunity granuloma were used. The same result is achieved by using human serum with a high antithyroid antibody titer obtained from patients with Hashimoto's disease. To obtain the antigenic material for the immunologic investigation, the cytologic aspiration method was used. In all those cases in which the aspirated material contained thyroid colloid, the result was positive, as observed in various types of goiter, papillary carcinoma and metastases from papillary carcinoma. By contrast, it was negative when neither colloid nor differentiated cells were present, as in undifferentiated carcinoma, lymphosarcoma, lymphomatous goiter with extensive replacement of parenchyma, thyroglossal cysts, and suppurative thyroiditis occurring in a pre-existing thyroid nodule. The method under consideration permits the diagnosis of metastases of papillary carcinoma before the pathologic examination of operative or biopsy material
