Injection of 5 ng of estradiol benzoate (Eb) into the third ventricle of ovariectomized rats resulted in a decline in plasma LH at 2 and 4 hr after the injection as determined by radioimmunoassay. Control injections of saline into the ventricle or of the same amount of Eb intravenously failed to alter plasma LH significantly. The responsiveness of the pituitary to LRF was not altered at either 2 or 4 hr following the intraventricular injection of estradiol. It is concluded that minute doses of estradiol can act on the central nervous system to inhibit LH release in the ovariectomized rat. No changes in plasma FSH followed these injections.
