Abstract
Rate equations and rate parameters are determined for the coupled-reaction assay for serum glutamic-oxaloacetic transaminase in which malic dehydrogenase is used. These are used to interpret observed curves of absorbance vs. time, to select optimum substrate concentrations, and to estimate nonrandom analytical errors. A simple, systematic, general approach is presented, which can readily be applied to other enzyme assays in which two or more reactions are coupled. It depends for its simplicity on access to an adequate computing facility.
