1 The Schwann cell lineage: embryonic and early postnatal development

Abstract
This chapter examines recent work on Schwann cell development. It discusses how Neu differentiation factor (NDF) might regulate entry of neural crest cells to the glial lineage, describes the recently identified rat Schwann cell precursor, and discusses the role of NDF in controlling both precursor survival and precursor maturation to generate immature Schwann cells. It reviews current knowledge about identified factors that are capable of regulating further differentiation of immature Schwann cells, in particular, expression of the myelin phenotype. This includes discussion of the cAMP model of Schwann cell differentiation in vitro, and the roles of fibroblast growth factors, insulin-like growth factors, and transforming growth factor-β in the positive and negative regulation of the expression of the major protein of Schwann cell myelin — protein zero. Finally, gene regulatory proteins in the Schwann cell lineage are reviewed.
