Abstract
Localization in green leaves of glycine decarboxylase and serine hydroxymethyltransferase (EC 2.1.2.1) was investigated. Subcellular preparations of green leaves were fractionated by non-linear sucrose isopicnic centrifugation of 2.5 to 1.3 M at 74,700 × or 1.8 to 0.6M at 11,800×g and by centrifugation in non-aqueous media. Glycine decarboxylase was located in mitochondria and serine hydroxymethyltransferase was principally located in mitochondria and partly in chloroplasts. Chloroplastic serine hydroxymethyltransferase is thought to be responsible for glycine formation from serine derived from photosynthesized 3-phosphoglycerate. A scheme for glycolate metabolism and photorespiration is presented.
