Background/Introduction: Cardiac remodeling (hypertrophy or dilation) is often associated with left ventricle (LV) dysfunction, a major risk for sudden cardiac death, claiming thousands of lives on a yearly basis. This owes to the poor understanding of the molecular cues involved in the development of cardiac remodeling and more importantly its associated contractile dysfunction. We have previously reported that SLMAP regulates the electrophysiological properties of the heart in vivo however, the role of SLMAP in cardiomyocyte contraction and its expression in pathological hearts were not as yet evaluated.
Purpose: To evaluate the expression of SLMAP in cardiac remodeling and its role in cardiomyocyte contraction.
Methods: We combined proteomics with western blot analysis to assess the expression of SLMAP in Spontaneously Hypertensive Rats SHR hearts. We also evaluated the interaction of SLMAP with striatin, a regulator of cardiomyocyte contraction. Confocal microscopy was employed to evaluate the contraction rate of cultured cardiomyocytes. Live imaging was used to assess the role of SLMAP in hypertrophy and adrenergic-induced calcium signaling in cardiomyocyte with manipulated expression of SLMAP (Knockdown-KD, and Overexpression-OE using adenoviral transduction particles).
