“Clostridium difficile: Methods and Protocols” is part of the Springer Methods in Molecular Biology book series. The book provides step-by-step protocols presented in a level of detail not possible in published manuscripts. These protocols are often written by the investigators who developed them and who are considered experts in their respective areas. Each chapter starts with a brief introduction followed by the research protocols, which consist a listing of the materials and reagents needed, detailed procedures, and notes that highlight tips and potential pitfalls when conducting the experiments.

This book is timely for 2 reasons. The first is the dramatic change in C. difficile epidemiology that has highlighted the general lack of knowledge about the organism at the molecular level and how C. difficile interacts with the host. The second is related to the first, in that until the preceding 10 years, reliable methods to genetically manipulate C. difficile have been lacking, thus hampering the ability to study the organism at the molecular level. However, our enthusiasm for this volume is slightly dampened because some of the methods described have been in use for well over 10 years, and alternate materials and methods, when available, are frequently not discussed.
